Mode of action: The enzyme is an alcohol dehydrogenase most active on n-butanol but with low activity on ethanol.

Unit Definition: One unit (U) of enzyme activity is defined as the absorbance measured after incubation for 10 min at 340nm (NAD+ turning into NADH) units (U) are defined as mmol/min of product released.

Adh131 Alcohol dehydrogenase activity vs. temperature

Temperature Optimum: The enzyme in relatively active at broad temperature range (50-65°C)with optimum around 60°C.

Adh131 Alcohol dehydrogenase activity vs. pHpH Optimum: pH range is about 8-9 with optimum around  pH 8,0.

 

Adh131 Alcohol dehydrogenase thermostabilityThermostability: The enzyme is stable over 400 minutes at temperature at 65°C.

Unit Assay Conditions: 

100 µl sample:

 

  • 100 µl 1-butanol (110mM)
  • 200 µl NAD+  (50mM)
  • 100 µl KPO4 pH 8 (500mM)
  • 10 µl zinksalt  (100mM)
  • 390 µl dH2O

 

Incubated at 60°C for 10min.  Reaction stopped on ice.  Spin shortly at 4°C before measuring the absorbance at 340nm.
