Mode of action: The enzyme is an alcohol dehydrogenase most active on n-butanol but with low activity on ethanol.
Unit Definition: One unit (U) of enzyme activity is defined as the absorbance measured after incubation for 10 min at 340nm (NAD+ turning into NADH) units (U) are defined as mmol/min of product released.
Temperature Optimum: The enzyme in relatively active at broad temperature range (50-65°C)with optimum around 60°C.
pH Optimum: pH range is about 8-9 with optimum around pH 8,0.
Thermostability: The enzyme is stable over 400 minutes at temperature at 65°C.
Unit Assay Conditions:
100 µl sample:
- 100 µl 1-butanol (110mM)
- 200 µl NAD+ (50mM)
- 100 µl KPO4 pH 8 (500mM)
- 10 µl zinksalt (100mM)
- 390 µl dH2O
Incubated at 60°C for 10min. Reaction stopped on ice. Spin shortly at 4°C before measuring the absorbance at 340nm.
