Mode of action: The enzyme hydrolyses terminal, non-reducing b-D-galactose residues in b-D-galactosides.
Unit Definition: One unit (U) is defined as the amount of enzyme which liberates 1 mmol of p-nitrophenol per min from pNP-b-galactoside, under the given assay conditions.
Unit Assay Conditions: The standard assay for cellulase activity was done by incubating the enzyme at 80°C for 10 min, with 1 % (w/v) pustulan, as a substrate in 0.1M sodium phosphate buffer, pH 5,5. The reducing sugars released were detected by the dinitrosalicylic acid method using glucose as standard.
Enzyme properties:
Temperature Optimum: The enzyme activity is limited below 65°C but increases with temperature up to 100°C
pH Optimum: pH range is about 4-7 with optimum around 6.5
Thermostability: The ß-galactosidase heat stability at 70,80 and 91°C
